Peptide Microarray Quality Control

Traditional peptide array and microarray manufacturing done in-situ generally limits the size of the peptide that can be put onto the microarray. Because in-situ synthesis directly onto glass to make microarrays is not very efficient, traditional in-situ synthesis manufacturing of microarrays has been limited to only 8 amino acids. The efficiency decreases as the length of amino acid sequence increases and once on the "chip" there is not a way to test the quality of the peptides without using the microarray. Spot synthesis is used by some of our competitors, which makes the peptide on membrane. With that method it is not possible to use mass spec to QC for the quality of the peptide eluted off those type of membranes. The membrane spot synthesis type macroarrays are not high performance as microarrays. With the Arrayit Peptide Microarray Platform the peptides are made ex-situ using efficient solid phase synthesis coupling technology, each peptide is quality checked by mass spectrometry and then they are mechanically printed into microarrays using US Patent 6,101,946. The peptides are covalently bound to the glass slide available for any type of binding study you wish to perform in a highly multiplex and low volume reaction. The microarray are compatible with all standard high sensitive fluorescent microarray scanners.
